NGAL Antigen (Cat# 21231215M)
Iron-traffickingprotein involved in multiple processes such as apoptosis, innate immunity andrenal development. Binds iron through association with 2,5-dihydroxy benzoicacid (2,5- DHBA), a siderophore thatshares structural similarities with bacterial enterobactin, and delivers orremoves iron from the cell, depending on the context. Iron-bound form (holo-24p3)is internalized following binding to the SLC22A17 (24p3R) receptor, leading torelease of iron and subsequent increase of intracellular iron concentration. Incontrast, association of the iron-free form (apo-24p3) with the SLC22A17(24p3R) receptor is followed by association with an intracellular siderophore,iron chelation and iron transfer to the extracellular medium, thereby reducingintracellular iron concentration. Involved in apoptosis due to interleukin-3(IL3) deprivation: iron-loaded form increases intracellular iron concentrationwithout promoting apoptosis, while iron-free form decreases intracellular ironlevels, inducing expression of the proapoptotic protein BCL2L11/BIM, resultingin apoptosis. Involved in innate immunity, possibly by sequestrating iron,leading to limit bacterial growth.
In the case of acute kidney injury (AKI), NGAL issecreted in high levels into the blood and urine within 2 hours of injury. BecauseNGAL is protease resistant and small, the protein is easily excreted anddetected in the urine. NGAL levels in patients with AKI have been associatedwith the severity of their prognosis and can be used as a biomarker for AKI.NGAL can also be used as an early diagnosis for procedures such as chronickidney disease, contrast induced nephropathy, and kidney transplant.
