Zika virus is a cousin of dengue fever and yellow fever virus in the mosquito-borne flavivirus family. Since the first report in Uganda in 1947 in monkeys, outbreaks of zika virus disease in human have been recorded in Africa, Asia, and the Americas. Zika virus infection has been reported to be associated with Guillain-Barre Syndrome and microcephaly.
Infection with Zika virus may be suspected based on symptoms and recent history of travel (e.g. residence in or travel to an area with active Zika virus transmission). A diagnosis of Zika virus infection can only be confirmed through laboratory tests on blood or other body fluids, such as urine, saliva or semen. In-direct ELISA for Zika NS1 Antigen and direct ELISA for Zika antigen stimulated antibodies are two powerful means to identify the case.
America Diagnostics’ AmeriDx® Zika NS1 Antigen ELISA Kit is a Sandwich Enzyme-Linked Immunosorbent Assay. A Zika NS1 specific mouse monoclonal antibody is first coated on a 96-well plate. Standards and samples are then added to the wells. The Zika NS1 antigen binds to the immobilized capture antibody. Next, a biotinylated mouse monoclonal anti-Zika NS1 detection antibody is added, producing an antibody-antigen-antibody “sandwich”. Avidin-horseradish peroxidase (Avidin-HRP) is subsequently added, followed by the addition of ELISA Substrate Solution, producing a blue color. The blue color intensity is proportional to the concentration of Zika NS1 antigen present in the sample. Finally, the addition of Stop solution changes the reaction color from blue to yellow, and the microwell absorbance is read at 450 nm and 650 nm with a microplate reader.
