Human NGAL (Neutrophil Gelatinase-Associated Lipocalin) is a potential new biomarker for the acute kidney injury (AKI).
Under normal conditions, NGAL levels are low in urine and plasma, but they rise sharply from basal levels in response to kidney injury and peaks within a very short time – as much as 24 hours or more before any significant rise in serum creatinine. The finding of a raised urinary or plasma level of NGAL cannot be independently diagnostic of any single pathology, since a variety of independent pathologies (e.g. inflammation, infection and certain types of cancer) are associated with raised levels of urinary or plasma NGAL.
America Diagnostics’ AmeriDx® NGAL Antigen ELISA Kit is a Sandwich Enzyme-Linked Immunosorbent Assay. A NGAL specific mouse monoclonal antibody is first coated on a 96-well plate. Standards and samples are then added to the wells. The NGAL antigen binds to the immobilized capture antibody. Next, a biotinylated mouse monoclonal anti-NGAL detection antibody is added, producing an antibody-antigen-antibody “sandwich”. Avidin-horseradish peroxidase (Avidin-HRP) is subsequently added, followed by the addition of ELISA Substrate Solution, producing a blue color. The blue color intensity is proportional to the concentration of NGAL antigen present in the sample. Finally, the addition of Stop solution changes the reaction color from blue to yellow, and the microwell absorbance is read at 450 nm and 650 nm with a microplate reader.
