Prostate-specific antigen (PSA) is a protein produced by normal prostate cells. This enzyme participates in the dissolution of the seminal fluid coagulum and plays an important role in fertility. The highest amounts of PSA are found in the seminal fluid; some PSA escapes the prostate and can be found in the serum.
Rising levels of PSA in serum are associated with prostate cancer. The PSA level also tends to rise in men with benign prostatic hyperplasia (BPH) and is a good marker for prostate volume. PSA levels are usually elevated in men with acute bacterial prostatitis.
America Diagnostics’ AmeriDx® ELISA Kit is a Sandwich Enzyme-Linked Immunosorbent Assay. A human PSA specific mouse monoclonal antibody is first coated on a 96-well plate. Standards, positive controls, negative controls, and samples are then added to the wells. The PSA binds to the immobilized capture antibody. Next, a biotinylated mouse monoclonal anti-human PSA detection antibody is added, producing an antibody-antigen-antibody “sandwich”. Avidin-horseradish peroxidase (Avidin-HRP) is subsequently added, followed by ELISA Substrate Solution, producing a blue color. The blue color intensity proportional to the concentration of PSA present in the sample. Finally, the addition of the Stop solution changes the reaction color from blue to yellow, and the microwell absorbance is read at 450 nm and 650 nm with a microplate reader.
